Omeprazole works by irreversibly blocking the proton pumps in your stomach, which are the primary producers of gastric acid. However, these pumps are most active after a period of fasting, in anticipation of a meal. This is the fundamental reason why you should not take omeprazole at night, as taking it before bed misses the optimal timing for it to be most effective.
